Abstract(s) :

(Anglais) Escherichia coli is one of most used host microorganisms in biotechnology for the production of a large variety of indus- trially relevant components, such as recombinant proteins, insulin and growth hormone. However, the production of acetate during the culture, a phenomenon which is known as ’Overflow metabolism’, inhibits the cell growth. Fed-batch mode is used in E. coli cultures because it allows to reduce acetate formation by manipulation of the feed-stream. An intuitive regulation strategy may be to control the acetate concentration in the culture media at a very low value. However, there is no acetate probe on the market allowing on-line measurements. In this work, state estimation of the acetate concentration of a BL21(DE3) E. coli strain is presented. For this purpose, an identified model is needed. Hence, pa- rameter identification of the mechanistic model of E. coli is carried out, as well.
